Application: WB: 1/5000 - 1/40000 IHC: 1/1600 - 1/3200 Flow: 1/200 - 1/1000 IF/ICC: 1/2000 - 1/10000 IP: 1/50 Protein Function Has both glyceraldehyde-3-phosphate dehydrogenase and nitrosylase activities, thereby playing a role in glycolysis and nuclear functions, respectively. Participates in nuclear events including transcription, RNA transport, DNA replication and apoptosis. Nuclear functions are probably due to the nitrosylase activity that mediates cysteine S-nitrosylation of nuclear target proteins such as SIRT1, HDAC2 and PRKDC (By similarity). Glyceraldehyde-3-phosphate dehydrogenase is a key enzyme in glycolysis that catalyzes the first step of the pathway by converting D-glyceraldehyde 3-phosphate (G3P) into 3-phospho-D-glyceroyl phosphate. Post-translational S-nitrosylation of Cys-152 leads to interaction with SIAH1, followed by translocation to the nucleus. ISGylated. |

Recombinant GAPDH Antibody (Ab104788) - IF
IF analysis of GAPDH (green) in HeLa cells. The cells were fixed and permeabilized with 100% methanol for 5 minutes, and blocked with 2% BSA for 1 hour at room temperature. Cells were stained Recombinant GAPDH Antibody (Ab104788) at 1/100 dilution in blocking buffer overnight at 4°C, and then incubated with Goat Anti-Rabbit IgG H&L (AF488) at a dilution of 1/1000 for 1 hour at room temperature in the dark (green). Cells were counterstained with DAPI (blue). Images were taken on the confocal laser scanning microscope.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of prostate cancer tissue labelling GAPDH with Recombinant GAPDH Antibody (Ab104788) at 1/1600. Heat mediated antigen retrieval was performed using Tris/EDTA buffer pH 9.0.
Recombinant GAPDH Antibody (Ab104788) - Flow Cytometry
Overlay histogram showing HeLa cells stained with Recombinant GAPDH Antibody (Ab104788) (Red). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% TritonX-100 for 15 min. The cells were then incubated in Recombinant GAPDH Antibody (Ab104788) (1/1000 dilution) in 1x PBS/1% BSA for 30 min at room temperature. The secondary antibody was used a Goat Anti-Rabbit AF488 (IgG H+L) at 1/2000 dilution for 20 min at room temperature. Unlabelled sample (Black) was used as a control.
