How to operate the reconstitution and preservation of recombinant protein?

1) During transportation, lyophilized powder will adhere to the pipe wall or pipe cover due to external forces. Before opening the cover, the lyophilized powder will be collected at the bottom of the pipe through centrifugal operation to avoid loss and waste. Rapid centrifugation before capping 10000-12000 rpm, 30s; If there is no high-speed centrifuge, centrifuge at 3000-3500 rpm for 5min.

2) After centrifugation, add the provided re-dissolving buffer into the lyophilized powder, gently blow and mix with the gun head, and resuspend until the concentration is not less than 100 μg/mL. Note: When sub-packed and frozen, the protein concentration shall not be less than 100 ug/mL, and the volume of each tube shall not be less than 20 μL.

3) The cytokine or recombinant protein solution directly dissolved by the redissolved buffer can be stored at 2-8 ℃ for up to 1 week. For the experiment with short cycle, the recombinant protein solution stored under this condition can be directly added into the culture system and used up within one week. If long-term storage is required, the solution containing carrier protein (0.1% BSA, 5% HAS, 10% FBS or 5% trehalose) needs to be further diluted, and then sub-packaged and frozen at - 20 ℃ to - 80 ℃.

4) Please avoid repeated freezing and thawing when storing the product. Repeated freeze-thaw and ice crystals will destroy the spatial structure of the recombinant protein, resulting in the denaturation of the protein to form a polymer and the conformation change of the recombinant protein, thus reducing the binding ability of the antibody and accelerating the degradation rate of the protein.
