生化机理 秋水仙碱是一种微管蛋白抑制剂,通过与微管蛋白结合(IC50 = 3.2 μM)可阻断微管的聚合。研究表明,秋水仙碱还能通过作用于组织纤溶酶原激活剂(tPA)和破坏细胞的细胞骨架来导致细胞凋亡。还有报告称,细胞暴露于秋水仙碱会导致细胞中 Ca2+ 浓度的改变,这也可能导致细胞凋亡。由于有丝分裂的速度加快与癌细胞增殖有关,用秋水仙碱阻断微管功能已被用作抗癌治疗的一种方法。例如,秋水仙碱可阻断 MCF-7 人类乳腺癌细胞的生长,其 IC50 值为 13 nM。秋水仙碱通过抑制中性粒细胞的运动和活性(在痛风性关节炎小鼠模型中为 5 μmol/kg)产生抗炎作用,并抑制尿酸沉积,这是治疗痛风的一个关键方面。

遇光变暗。易溶于乙醇、氯仿,溶于水、乙醚、苯,不溶于石油醚;溶解度:45g/L (20°C)、1g/220ml 乙醚、1g/100ml 苯。pK(20℃)=12.35,pH值5.9。因其氮原子在侧链上是酰胺结构,故不呈碱性。

Colchicine, a tubulin Inhibitor, has been shown to block polymerization of microtubules by binding to tubulin. Studies indicate that this reagent also causes apoptosis of cells by acting on tissue plasminogen activators (tPA) and disrupting the cytoskeleton of cells. Exposure of cells to Colchicine has also been reported to result in alterations of Ca2+ concentrations in cells which may also result in apoptosis.
