This molecule consists of a tetrapeptide sequence DEVD (a specific recognition sequence for Caspase-3/7) and a fluorophore AMC (7-amino-4-methylcoumarin). Under normal circumstances, AMC is blocked by the DEVD sequence and does not emit light; when Caspase-3 or -7 is activated, it will recognize and cleave the bond between DEVD and AMC, releasing free AMC molecules, which then emit detectable blue fluorescence (Ex/Em ≈ 350/450 nm). |
