生化机理 盐酸洛哌丁胺是一种阿片类受体激动剂,对 MOR(μ-阿片受体)具有选择性。这种对μ-阿片受体的特异性和对大脑的有限渗透性使得洛哌丁胺无法表现出阿片类药物通常具有的中枢性全身效应,而是表现出对痛觉感受器的局部作用和由此产生的抗过敏作用。洛哌丁胺可通过高电压激活的钙通道对 Ca2+ 通量产生影响,以应对高浓度的细胞外 K+。对阿片受体亚型 μ、δ 和 κ 的 K i 值分别为 3、48 和 1156 nM。高剂量可激活中枢μ受体。止泻、止痒和抗过敏药。
