生化机理 阿昔洛韦是一种抗病毒化合物,可用于诱导转染有单纯疱疹病毒胸苷激酶(HSV-TK)的细胞凋亡。该化合物可通过改变白血病细胞系(如表达 p185BCR-ABL 的 OM9;22 细胞)的细胞周期,使 G0/G1 期同步,从而发挥抑制作用。无环鸟苷三磷酸通过 HSV-TK 磷酸化形成,可抑制病毒 DNA 聚合酶。阿昔洛韦还能抑制由 v-abl 或 bcr-abl 转化的成纤维细胞 NIH 3T3 的增殖。鸟苷衍生物能竞争性地抑制病毒 DNA 聚合酶。对单纯疱疹 1 和 2(EC 50 值分别为 0.04 和 0.44 μM)以及水痘-带状疱疹病毒(IC 50 = 0.22 μM)具有高度特异性。
