链霉抗生物素蛋白是一个非糖基化的蛋白质。 它对生物素具有优异的高亲和力,并且在大幅度的pH和温度范围内稳定。 由于其对生物素的高亲和力,它被用来桥接生物素化的分子。 每个链霉亲和素分子可以结合4个生物素分子。链霉亲和素是一种来源于阿维丁链霉菌(streptomyces avidinii)的生物素结合蛋白质,其非特异性结合远比亲和素低,链霉亲和素蛋白具有在中性pH值几乎没有净电荷,不含有碳水化合物等更有利的化学性质,所以它被广泛用来作为抗生物素蛋白的替代品。 在结构上,链霉亲和素含SA 的127AA 最佳核心结构,以同源四聚体的形式存在,每摩尔的四聚体分子可结合四摩尔的生物素分子,国际标准比活性12-15 IU/mg。该特性联合生物素偶联抗原/抗体检测技术,使其广泛应用于免疫学中ELISA检测信号的放大。此外,本品主要可用于免疫微孔板、免疫层析硝酸纤维素膜,基因芯片、蛋白芯片的表面包被,快速固定生物素化的抗原、抗体、核酸等物质。也可用于偶联NHS 磁性磁珠、树脂微球,作为发光诊断、生物素化物质分离、磁性试纸条免疫层析等方面的应用。高亲和力生物素结合蛋白(Kd 约为10-15),其在宽的pH和温度范围内可保持稳定。每个链霉亲和素分子可结合4个生物素分子。链霉亲和素缺乏存在于亲和素上的碳水化合物侧链,并具有接近中性的等电点(pI为5.5-6.5,亲和素为10)。因此,链霉亲和素通常显示出比亲和素更低的非特异性结合水平。1%溶液在280 nm处的消光系数为32。高亲和力的生物素结合蛋白。

Streptavidin from Streptomyces avidinii has been used: in the pre-functionalization of self-propelled catalytic micromotors in the fabrication and modification of hydroxyapatite-chitosan (HA-CTS) nanofilm-coated substrates as an analyte to bind biomolecules to the gold-inverted polymer solar cells (Au-IPSC)

Streptavidin is used as a tetrameric protein used to visualize biotin conjugated molecules in ELISA. Due to the strong affinity of biotin and biotinylated molecules has made streptavidin one of the most important components in diagnostics and laboratory kits. Streptavidin is widely used in molecular biology through its unique high affinity for the biotin.
