Bexarotene

Bexarotene(Aladdin B125743 is a retinoid compound with high specificity for retinoid  X receptors (RXRs). It is approved as a chemotherapeutic for the treatment of cutaneous T-cell lymphoma and has also been investigated for its potential therapeutic role in Alzheimer’s disease.

The link between amyloid-β plaque accumulation and Alzheimer’s disease is well documented. Misfolded amyloid-β aggregates disrupt neuronal signaling by blocking the transmission of electrical and chemical messages within the brain. These peptides are derived from the cleavage of amyloid precursor protein (APP), a transmembrane glycoprotein. While the exact physiological function of amyloid-β remains unclear, its aggregation into plaques is a hallmark associated with the progression of Alzheimer’s disease.

Apolipoprotein E (ApoE) is a protein responsible for promoting the clearance of amyloid-β deposits. Its transcription is normally triggered by nuclear receptor activity, including RXR activation. Acting as an RXR agonist, bexarotene significantly increases soluble amyloid-β clearance within hours in animal models of Alzheimer’s disease. Within three days, plaque burden can be reduced by over 50%. Additionally, bexarotene has been shown to help reverse cognitive impairments and enhance neural circuit performance.

Ongoing studies continue to explore bexarotene’s effects in Alzheimer’s disease models and its potential in other research areas. Since RXR activation engages the body’s natural amyloid-β clearance pathways, bexarotene—along with other retinoic acid receptor (RAR) agonists such as 9-cis-Retinoic Acid (AladdinC134191) and 13-cis-Retinoic Acid (Aladdin I129856)—may hold increasing significance for investigations into neurodegenerative disorders.
