生化机理	Epitope tags are useful for the labeling and detection of proteins using immunoblotting, immunoprecipitation, and immunostaining techniques. Because of their small size, they are unlikely to affect the tagged protein’s biochemical properties. DYKDDDDK tag, also known as FLAG tag, is a short peptide sequence that is commonly used as a protein tag to facilitate protein purification, detection, and localization. The DYKDDDDK tag is widely used in molecular biology and biochemistry research, and has been employed in various applications, such as protein-protein interaction studies, protein expression analysis, and protein localization studies. The FLAG tag structure has been optimized for compatibility with the proteins it is attached to, in that it is more hydrophilic than other common epitope tags and therefore less likely to denature or inactivate proteins to which it is appended.

IF analysis of DYKDDDDK tag (red) in HEK293-EGFP-Flag Tag cells. The cells were fixed and permeabilized with 100% methanol for 5 minutes, and blocked with 2% BSA for 1 hour at room temperature. Cells were stained with Recombinant DYKDDDDK tag (Binds to Flag tag) Antibody (Ab224136) at 2.0 μg/mL in blocking buffer overnight at 4°C, and then incubated with Goat Anti-Mouse IgG H&L (AF647) (Ready to use) (Ab218689) at a dilution of 1/1000 for 1 hour at room temperature in the dark (red). PBS instead of the primary antibody was used as the secondary antibody only control. Cells were counterstained with DAPI (blue), and the EGFP protein is green. Images were taken on the confocal laser scanning microscope.
