生化机理	Human influenza hemagglutinin (HA) is a surface glycoprotein required for the infectivity of the human virus. The HA tag is derived from the HA molecule corresponding to amino acids 98-106 has been extensively used as a general epitope tag in expression vectors. Many recombinant proteins have been engineered to express the HA tag, which does not appear to interfere with the bioactivity or the biodistribution of the recombinant protein. This tag facilitates the detection, isolation, and purification of the proteins.

IF analysis of HA tag (Red) in HEK293T cells transfected with a eGFP-HA tag. The cells were fixed with 4% Paraformaldehyde Fix Solution (P395744) for 15 minutes, permeabilized with 0.3% Triton X-100 (I743383) for 15 minutes, and blocked with 5% BSA for 1 hour at room temperature. Cells were stained with HA tag Mouse mAb (TRITC) (Ab222050) at 1/1000 dilution in blocking buffer for 2 hour at room temperature in the dark. Cells were counterstained with DAPI (Blue). Images were taken on the confocal laser scanning microscope.

Flow Cytometry analysis of HA tag expression by HEK293T cells transfected with a eGFP-HA tag (red) compared to an isotype control (blue). The cells were fixed with 4% Paraformaldehyde Fix Solution (P395744) for 15 minutes, permeabilized with 0.3% Triton X-100 (I743383) for 15 minutes and then stained with HA tag Mouse mAb (TRITC) (Ab222050) at a dilution of 1/500 for 30 minutes at 4°C in the dark. Blue - Mouse IgG (TRITC) (Ab225743).
