生化机理	Epitope tags are useful for the labeling and detection of proteins using immunoblotting, immunoprecipitation, and immunostaining techniques. Because of their small size, they are unlikely to affect the tagged protein’s biochemical properties. DYKDDDDK tag, also known as FLAG tag, is a short peptide sequence that is commonly used as a protein tag to facilitate protein purification, detection, and localization. The DYKDDDDK tag is widely used in molecular biology and biochemistry research, and has been employed in various applications, such as protein-protein interaction studies, protein expression analysis, and protein localization studies. The FLAG tag structure has been optimized for compatibility with the proteins it is attached to, in that it is more hydrophilic than other common epitope tags and therefore less likely to denature or inactivate proteins to which it is appended.

IF analysis of DYKDDDDK tag (Binds to Flag tag) (Red) in HEK293T cells transfected with a EGFP-DYKDDDDK. The cells were fixed with 4% Paraformaldehyde Fix Solution (P395744) for 15 minutes, permeabilized with 0.3% Triton X-100 (I743383) for 15 minutes, and blocked with 2% BSA for 1 hour at room temperature. Cells were stained with Recombinant DYKDDDDK tag (Binds to Flag tag) Antibody (Biotin) (Ab218446) at 1/1000 dilution in blocking buffer for 2 hour at RT and then incubated with Recombinant Streptavidin Protein (Cy5) (rp169047) at a dilution of 1/1000 for 1 hour at room temperature in the dark. Cells were counterstained with DAPI (Blue). Images were taken on the confocal laser scanning microscope.

Flow Cytometry analysis of DYKDDDDK tag (Binds to Flag tag) expression by HEK293T cells transfected with a EGFP-DYKDDDDK tag (red) compared to an isotype control (blue). The cells were fixed with 4% Paraformaldehyde Fix Solution (P395744) for 15 minutes, permeabilized with 0.3% Triton X-100 (I743383) for 15 minutes and then stained with Recombinant DYKDDDDK tag (Binds to Flag tag) Antibody (Biotin) (Ab218446) at a dilution of 1/900 for 30 minutes at 4°C in the dark. Then incubated with Recombinant Streptavidin Protein (APC) (rp169048). Blue - Mouse IgG (Biotin) (Ab180306). Black - Unlabelled control, cells without incubation with primary antibody.
