Product Description

Proliferating cell nuclear antigen (PCNA) is a member of the DNA sliding clamp family of proteins that assist in DNA replication. Crystal structure data suggests that a PCNA homotrimer ring can encircle and slide along the DNA double helix. Multiple proteins involved in DNA replication, DNA repair, and cell cycle control bind to PCNA rather than directly associating with DNA, thus facilitating fast processing of DNA. PCNA protein expression is a well-accepted marker of proliferation.

PCNA Mouse mAb (Ab155826) - ICC/IF
IF analysis of PCNA (green) in HeLa cells. The cells were fixed and permeabilized with 100% methanol for 5 minutes, and blocked with 2% BSA for 1 hour at room temperature. Cells were stained with PCNA Mouse mAb (Ab155826) at 1/2000 dilution in blocking buffer overnight at 4°C, and then incubated with Goat Anti-Mouse IgG H&L (AF488) at a dilution of 1/1000 for 1 hour at room temperature in the dark (green). Cells were counterstained with DAPI (blue). Images were taken on the confocal laser scanning microscope.

Flow analysis of PCNA (red line) in HeLa cells. The cells were fixed and permeabilized with 100% methanol for 5 minutes. PCNA Mouse mAb (Ab155826) at 1/4000 dilution for 1 hour at 4°C, and then incubated with Goat Anti-Mouse IgG H&L (FITC) (Ab156254) at a dilution of 1/1000 for 30 minutes at 4°C in the dark . Unlabeled sample (black line) was used as a negative control.
