Its main ingredients are SDS, DTT, bromophenol blue, buffer salt solution, etc. SDS can combine with protein to make the protein SDS complex carry a large number of negative charges. At this time, the charge of the protein itself is completely covered by SDS, eliminating the difference in the charge of various proteins; SDS can also break the intramolecular and intermolecular hydrogen bonds and destroy the secondary and tertiary structures of protein molecules. DTT can break the disulfide bond between cysteine residues, destroy the protein structure, and eliminate the differences between protein structures. The electrophoretic migration of proteins (subunits) with no charge and structural differences is only related to their molecular weight. Bromophenol blue is used as an indicator for electrophoresis, which can roughly indicate the end time of electrophoresis. usage method: 1. Dilute the protein sample and the sample buffer solution at a ratio of 3:1 for use. If the concentration of protein sample is too high, it can be diluted with Grade III water or Grade I water. 2. After mixing, heat it in 100 ℃ water bath (or metal bath) for 5 minutes to denature the protein. 3. After cooling to room temperature, centrifugate at 10000-14000 rpm for 2-5 minutes, and then directly load samples for electrophoresis. 4. After protein denaturation, if the protein is stored at room temperature for a long time or stored at low temperature, protein renaturation will occur, and it needs to be reheated for denaturation before electrophoresis.
